Mechanism of Action
Nefazodone functions as a post-synaptic 5-HT2A Receptor Antagonist, and at the same time, functions as a Serotonin Reuptake Inhibitor. This means that it is a novel, dual-action Serotonergic Antidepressant. Nefazodone's potent 5-HT2A receptor antagonism in combination with 5-HT reuptake inhibition appears to enhance 5-HT1A-mediated neurotransmission. Nefazodone has very selective serotonergic effects, with negligible affinity for cholinergic and histamine receptors and low affinity for alpha 1-adrenergic receptors.
